Semax increases BDNF expression in the rat hippocampus and cerebral cortex

This study investigated the neurotrophic effects of Semax, a heptapeptide ACTH analogue. Administration increased BDNF and NGF expression in the hippocampus and cortex, suggesting a molecular basis for its observed cognitive effects in prior research. The effects were region-specific and time-dependent.

Why this research is interesting

Semax is used clinically in Russia but is less well-known internationally. This study provides molecular-level evidence for its neurotrophic effects, helping explain the cognitive benefits reported in clinical use there.

What the research says

Semax administration in rats increased expression of BDNF (brain-derived neurotrophic factor) and NGF (nerve growth factor) in specific brain regions. These neurotrophins are associated with synaptic plasticity, learning, and memory.

What the evidence shows

This is an animal study measuring gene expression changes. It demonstrates a molecular effect but does not directly measure cognitive outcomes. Clinical data from human use in Russia exists but is not included in this study.

What remains uncertain

Whether BDNF increases in rat brain translate to cognitive benefits in humans is not established by this study alone. The clinical evidence from Russian studies has not been widely replicated in Western trial designs.
