FOXO4-DRI

Senolytic Peptide — Selective Apoptosis of Senescent Cells

Molecular Data

Formula
C168H258N44O49S
Molecular Weight
3856.1 g/mol

01

Overview

FOXO4-DRI is a synthetic D-retro-inverso peptide designed to disrupt the interaction between FOXO4 and p53 in senescent cells. Developed at the Erasmus MC Medical Center in Rotterdam, it represents a senolytic approach that selectively induces apoptosis in cells that have entered replicative senescence while sparing healthy proliferating cells.

02

Mechanism of Action

In senescent cells, FOXO4 binds p53 and sequesters it in the nucleus, preventing apoptosis. FOXO4-DRI displaces p53 from FOXO4 by competing for the FOXO4 binding interface, freeing p53 to translocate to mitochondria and initiate apoptosis. Because only senescent cells accumulate FOXO4, healthy cells are unaffected.
